Introduction
Based on similar principle as E.Z.N.A.® Viral RNA kit, E-Z® 96 Viral RNA kit is designed for automated high throughput isolation of Viral RNA from cell free fluids such as plasma, serum, urine, cell culture supernatants. The procedure completely removes contaminants and enzyme inhibitors making viral RNA isolation fast, convenient, and reliable. This kit has been verified to isolate viral nucleic acids from Hepatitis A and C and HIV. The kit is also suitable for isolation of total RNA from cultures cells, tissues, and bacteria.
Principle
The E-Z® Viral RNA Kits use the reversible binding properties of HiBind® matrix, a new silica-based, time-saving spin technology material. Combined with the speed of 96-well spin or vacuum manifold technology, up to 192 samples can be processed at the same time. It combines the reversible nucleic acid-binding properties of the HiBind™ matrix with the speed and versatility of the E-Z 96™ RNA plate to eliminate polysaccharides, phenolic compounds, and enzyme inhibitors from tissue lysates.
Procedure
The samples are lysed under highly denaturing buffer conditions to inactivate RNases, thus protecting RNA from degradation. After adjusting the binding conditions, the samples are loaded to the E-Z 96® RNA Plate. With a brief centrifugation or vacuum, the samples pass through the plate binding the RNA to the Hibind® matrix. After two wash steps, purified RNA will be eluted with RNase-free water.
