The Agarose Gel Extraction Kit uses HiBind^® technology to recover DNA fragments from 50 bp-40 kb from all grades of agarose gel in yields exceeding 85%. Each HiBind^® extraction column has a total capacity of 25-30 µg DNA. HiBind^® matrix avidly, but reversibly, binds DNA under certain optimal conditions. DNA is then easily eluted with de-ionized water or low salt buffer. The product is suitable for ligations, PCR sequencing, restriction digestion, and labeling reactions. The use of mini-columns facilitate the binding, washing, and elution steps thus enabling multiple samples to be simultaneously processed. Procedure

Perform agarose gel/ethidium bromide electrophoresis to fractionate DNA fragments. Carefully excise the fragment of interest while visualizing under UV light. Assuming a density of 1 g ml-1 of gel, add 1ml of Binding Buffer per gram of gel. Incubate to dissolve agarose and apply the DNA/agarose solution to a HiBind^® DNA spin-column and centrifuge. Discard the flow through and wash with SPW buffer diluted with ethanol to remove impurities. Centrifuge empty column for an additional minute to dry the column. Elute the DNA with sterile deionized water or TE buffer for further use.
