Introduction

The E.Z.N.A® Mag-Bind® Cycle-Pure Kit is designed to remove impurities such as excess nucleotides, primers, and small, non-targeted amplification products such as primer dimers from PCR products, while maintaining a high yield. The system is ideally suited for fully automated purification of PCR samples.

Principle

This novel system uses UBI’s Mag-Bind® Paramagnetic Particles to purify PCR products >100bp in size. DNA fragments as large as 20kb can be purified. PCR products bind to Mag-Bind® Paramagnetic Particles in the presence of Mag-Bind® Binding Buffer and remain tightly bound during washing. The purified PCR fragments are eluted in water and can be used for microarray, automated fluorescent DNA sequencing, restriction digestion and other applications.

Procedure

PCR products are first mixed with a Mag-Bind® Particles Solution/MGC Binding Buffer/absolute ethanol master mix. PCR products then selectively bind to the Mag-Bind® particles. With just two rapid wash steps, trace contaminants such as nucleotides, primers and small, non-targeted amplification products are removed. Pure DNA is eluted in Elution Buffer or water. Purified DNA can be directly used in downstream applications without the need for further purification.
