This method uses HiBind^® technology to recover DNA fragments ranging from 50 bp-40 kb, free of oligonucleotides, nucleotides, and polymerase in yields exceeding 85%. Binding conditions are adjusted by the addition of a specially formulated buffer, and the sample is applied to a HiBind^® DNA spin-column. HiBind™ matrix avidly, but reversibly, binds DNA under specific conditions allowing proteins and other contaminants to be removed by wash steps. Once impurities are removed DNA is eluted with deionized water (or low salt buffer) and is ready for other applications. The kit can also be used to purify DNA from any other enzymatic reaction. The use of mini-columns facilitate the binding, washing, and elution steps thus enabling multiple samples to be simultaneously processed. Procedure

Add equal volumes of buffer XP1 and enzymatic reaction and vortex to mix it. Apply the sample to HiBind^® DNA spin-column and wash the column twice with SPW Buffer diluted with absolute ethanol. Dry the column matrix with an additional spin step and elute DNA using sterile de-ionized water or TE buffer.
