By adapting the HiBind^® technology with 96 well plates, the E-Z 96^® Tissue DNA Kit provides a high-throughput method to purify genomic DNA from up to 30 mg tissue or two 0.6 cm of mouse tail in a 96 well plate format. The newly designed E-Z 96^® DNA Binding Plate has a binding capacity of 100 μg. The streamlined E.Z.N.A.^® procedure and convenient 96- well format allows processing of 96 samples in less than 2 hours. And the E-Z 96^® system is designed and optimized for use with vacuum manifolds and is compatible with most pipetting machines.

Principle

The E-Z 96^® Tissue DNA Kit combines the reversible nucleic acid-binding properties of the HiBind™ matrix with the speed and versatility of the E-Z 96™ DNA plate to eliminate polysaccharides, phenolic compounds, and enzyme inhibitors from tissue lysates.

Procedure

Tissue or tail samples are cut into small pieces, and then lysed in a specially formulated buffer containing proprietary detergent solution with proteinase. Binding conditions are then adjusted and the sample is applied to a E-Z 96^® DNA plate. Two rapid wash steps remove trace contaminants such as residual polysaccharides, and pure DNA is eluted in water or low ionic strength buffer. Purified DNA can be directly used in downstream applications without the need for further purification.
